No, the size of the image written to the card shouldn't have an effect on speed of use, far as I understand it.
Size of the card shouldn't matter - only thing that would happen there is that if you haven't resized the filesystem you can only access 8GB of your 64GB card, making it an expensive 8GB card.
More important is the class of the card - should be a class 10 if you want decent performance, as you want the higher read and write speeds.
